Clathrin: the molecular shape shifter

چکیده
Clathrin is best known for its contribution to clathrin-mediated endocytosis yet it also participates a diverse range of cellular functions. Key this clathrin's ability assemble into polyhedral lattices that include curved football or basket shapes, flat even tubular structures. In review, we discuss clathrin structure and coated vesicle formation, how utilised within different processes including synaptic recycling, hormone desensitisation, spermiogenesis, cell migration mitosis, remarkable ‘shapeshifting’ form lattice structures might contribute multiple

Molecular mechanisms of clathrin-independent endocytosis.
There is good evidence that, in addition to the canonical clathrin-associated endocytic machinery, mammalian cells possess multiple sets of proteins that are capable of mediating the formation of endocytic vesicles. The identity, mechanistic properties and function of these clathrin-independent endocytic pathways are currently under investigation. This Commentary briefly recounts how the field ...
متن کاملWeak Molecular Interactions in Clathrin-Mediated Endocytosis
Clathrin-mediated endocytosis is a process by which specific molecules are internalized from the cell periphery for delivery to early endosomes. The key stages in this step-wise process, from the starting point of cargo recognition, to the later stage of assembly of the clathrin coat, are dependent on weak interactions between a large network of proteins. This review discusses the structural an...
متن کاملClathrin triskelia show evidence of molecular flexibility.
The clathrin triskelion, which is a three-legged pinwheel-shaped heteropolymer, is a major component in the protein coats of certain post-Golgi and endocytic vesicles. At low pH, or at physiological pH in the presence of assembly proteins, triskelia will self-assemble to form a closed clathrin cage, or "basket". Recent static light scattering and dynamic light scattering studies of triskelia in...
متن کاملThe role of molecular chaperones in clathrin mediated vesicular trafficking
The discovery that the 70 kD "uncoating ATPase," which removes clathrin coats from vesicles after endocytosis, is the constitutively expressed Hsc70 chaperone was a surprise. Subsequent work, however, revealed that uncoating is an archetypal Hsp70 reaction: the cochaperone auxilin, which contains a clathrin binding domain and an Hsc70 binding J domain, recruits Hsc70(*)ATP to the coat and, conc...
متن کاملMolecular Shape Descriptors . 1 . Three - Dimensional Molecular Shape Descriptor
The paper presents and illustrates a method which uses numerical integration of the van der Waals envelope(s) to calculate with desired accuracy the molecular van der Waals volume and the three-dimensional molecular shape descriptor defined as the twin-number [OV(a, /?); NOV(/?, a)], where OV and NOV represent the overlapping and, respectively, the nonoverlapping van der Waals volumes of the mo...
